Histochemical Study on Changes of Alkaline Phosphatase Actvity, RNA and DNA of the Adrenal Cortex in Relation to Acceleration of Blood Coagulation in Vivo


Abstract
It was postulated by Kim and coworkers in our laboratory that acceleration of blood coagulation in vivo following infusion of Salt-Solutions, ACTH, and exposure of Electric Shock and Hypoxia, was due to increase of serum lipids(cholesterol and phospholipids) which were liberated from the adrenal cortex, and there were two routes inducing acceleration by those agents, that was either pituitary-adrenocortieal axis or from adrenal. Ro in our laboratory confirmed this difference morphologically on the adrenal cortex by changes of mitochondria. He however used only CaCls and ACTH as hemostatic agents. To gain more access to the facts, author chose 4 agents such as CaCJ2, Electric Shock, Hypoxa and ACTH, and studied the adrenal cortex histochemically.
Part I
To confirm the accelerating effects and mechanism of the hemostatic agents, author has made the, determinatin of clotting time respectively in normal, adrenalectomized and hypophysectomized rabbits after , infusion of CaChSoludion, ACTH, and exposure of Electric Shock and Hypoxia. The Results were as Follows,
1. CaCIQ, Electric Shock, Hypoxia and ACTH induce commonly marked shortening of blood clotting time in normal rabbits.
2. But their effects are all inhibited in adrenalectomized rabbits.
3. In hypophysectomized rabbits, CaCl2, Electric Shock are able to induce acceleration of blood coagulation, but Hypoxia .is not.
Part II
Histological and histochemical investigations were done by H-E staining, Sudan I for lipids, Schulz test for cholesterol, Gomori¢¥s Ca-Co method for Alkaline Phosphatase Activity, Pyroin-Methylgreen for RNA and Schiff¢¥s Reagent for DNA, on the adrenals from normal and hypophysectomized rabbits treated with those above mentioned agents.
The Results were as Follows
1. All 4 Stimuli that cause acceleration of blood coagulation are accompanied
with marked increase of Alkaline Phosphatase Activity and RNA in the zona fasciculata of the adrenals from normal rabbits.
2. The increase of RNA and Alkaline Phosphase Activity in nucleoli is most prominent in the cases treated with ACTH and less in Hypoxia, Elletric Shock, and not observed in CaCI2.
3. It seems to be possible by prominent increase of Alkaline Phosphatase Activity and RNA in nucleoli to differentiate whether the stimuli induce the changes of the adrenal cortex through pituitary or not.
